چکیده
Purpose The aim of this study was to monitor carbohydrate metabolism alongside cardiac structure and function, throughout HF progression. Methods Dilated cardiomyopathy (DCM) was induced in pigs (n = 4) by rapid ventricular pacing at 188 bpm for 4-5 weeks. Pigs were examined at baseline and at weekly time points throughout DCM progression. At each time point, cine MRI was used to assess cardiac structure and function, 0.05 mmol/kg hyperpolarised C2-pyruvate was administered intravenously and MRS was used to assess Krebs cycle-mediated C-glutamate production, and hyperpolarised C1-pyruvate was administered to assess H CO3 -

Hyperpolarised [2-C]Pyruvate Uniquely Reveals the Role of Acetylcarnitine as a Mitochondrial Substrate Buffer in the Heart
Introduction In normal hearts, Krebs cycle flux is maintained by continuous citrate synthase-mediated condensation of acetyl-CoA with oxaloacetate. In turn, acetylCoA is produced from the oxidation of fatty acids, ketone bodies or glucose, via glycolysis and the enzyme pyruvate dehydrogenase (PDH). Hyperpolarised [2C]pyruvate has been used in the isolated heart to observe the relationship betwe...
متن کاملIndexing tricarboxylic acid cycle flux in intact hearts by carbon-13 nuclear magnetic resonance.
Although the tricarboxylic acid (TCA) cycle is the prime means of carbon metabolism for energy generation in normal myocardium, the noninvasive quantification of TCA cycle flux in intact cardiac tissues is difficult. A novel approach for estimating citric acid cycle flux using 13C nuclear magnetic resonance (NMR) is presented and evaluated experimentally by comparison with measured myocardial o...
متن کاملDecreased contractile and metabolic reserve in peroxisome proliferator-activated receptor-alpha-null hearts can be rescued by increasing glucose transport and utilization.
BACKGROUND Downregulation of peroxisome proliferator-activated receptor-alpha (PPARalpha) in hypertrophied and failing hearts leads to the reappearance of the fetal metabolic pattern, ie, decreased fatty acid oxidation and increased reliance on carbohydrates. Here, we sought to elucidate the functional significance of this shift in substrate preference. METHODS AND RESULTS We assessed contrac...
متن کاملRates of glycolysis and glycogenolysis during ischemia in glucose-insulin-potassium-treated perfused hearts: A 13C, 31P nuclear magnetic resonance study.
The effects of 11.7 mM glucose, insulin, and potassium (GIK) on metabolism during ischemia were investigated in the perfused guinea pig heart using magnetic resonance spectroscopy. Intracellular metabolites, primarily glycogen and glutamate, were labeled with 13C by addition of [1-13C]glucose to the perfusate during a normoxic, preischemic period. 13C and 31P NMR spectroscopy was used to observ...
متن کاملHyperpolarized 13C Metabolic MRI of the Human Heart
RATIONALE Altered cardiac energetics is known to play an important role in the progression toward heart failure. A noninvasive method for imaging metabolic markers that could be used in longitudinal studies would be useful for understanding therapeutic approaches that target metabolism. OBJECTIVE To demonstrate the first hyperpolarized 13C metabolic magnetic resonance imaging of the human hea...
